Family Planning Program in Indonesia implemented through contraceptive methods that aim to improve training, participation and independence as well as reproductive health. The type of injectable contraception currently widely used is Depo Medroxyprogesterone Acetate (DMPA) which contains the progestin hormone, a synthetic material from progesterone. The use of DMPA causes hypoestrogen (estradiol) in the body. One of the roles of estradiol is as an antioxidant. Exogenous antioxidants can be obtained from green tea kombucha. Research method: Randomized posttest only control group research was conducted on female wistar rats (Rattus norvegicus) aged 2-3 months with a body weight of 150-200 grams. The rats were injected with DMPA once a week for 4 weeks. Thirty rats were randomly divided into negative and positive control, and three treatment groups given 0.5 ml, 1 ml, and 2 ml of green tea kombucha every day for 30 days. The results of the study showed that there was a difference in the average level of estradiol hormone after administration of green tea kombucha and DMPA injection. In conclusion, green tea kombucha is able to increase estradiol levels after hypoestrogen occurs after injecting DMPA contraception. The role of green tea kombucha as a phytoestrogen can be specifically studied in further studies
